Sample Preparation and RNA Extraction (PDF)
We recommend contacting us before sample preparation to obtain a quote. Once your samples are ready, please refer to the Services and Order tab and place an order via UC Stratocore.
Samples can be dropped off during business days from 9:30 AM to 4:00 PM at the following address:160 Panzeca Way, Kettering Building, Room 336, Cincinnati, OH 45267. Please enter through the main entrance. This is also our meeting and shipping address.
Please refer to the Directions & FAQs tab for guidance on sample preparation and RNA extraction. We also recommend contacting us in advance regarding sample collection, storage, and RNA/DNA extraction. We provide DNA and RNA extraction services as well.
RNA from fresh blood samples can be extracted directly without pre-treatment. However, if blood samples are stored for later RNA extraction, they must be treated according to established protocols. There are multiple approaches for proper blood sample preservation. Please contact GES Core for more information.
Yes. For degraded RNA samples, we use alternative approaches for RNA-seq and miRNA-seq. However, the cost of RNA-seq is higher compared to our standard protocols that use high-quality RNA for polyA RNA-seq.
If your RNA extraction protocol includes a phase separation step, ensure that the interphase (which contains genomic DNA) is not disturbed when transferring the aqueous phase. If sufficient RNA is available, it is advisable to leave a small portion of the aqueous phase behind.
For polyA RNA-seq using high-quality total RNA, minor genomic DNA carryover typically does not affect data quality, and DNase I treatment is not required. However, for rRNA/globin depletion RNA-seq, DNase I treatment during RNA extraction is required.
While we aim to provide a short turnaround time, several factors can affect it. For standard bulk sequencing (e.g., PE 2×61 bp), runs are typically scheduled at the end of each month. Other sequencing configurations (e.g., PE 2×151 bp) or single-cell sequencing may take longer. Please contact us for the current turnaround time and upcoming sequencing schedule.
The payment methods are listed in the invoice.
For simple services such as Bioanalyzer QC, a fixed rate is provided. For more complex services such as sequencing, the quote includes two components: a fixed service rate listed in UC Stratocore, which covers labor, and a pass-through cost that covers kits, consumables, etc., which is not listed. For large projects, the pass-through cost may be reduced. Please contact us for a final quote.
Yes. We provide supporting letters, service quotes, and standard Materials & Methods text for grant applications and manuscript preparation. We also offer consultation on experimental design and sample preparation. These services are free of charge.
Yes, we are open to collaboration. With our expertise, we welcome opportunities for joint grant applications and manuscript submissions. Publications resulting from our collaboration may be referenced to other users when similar services are requested.
